Dexmedetomidine Promotes Tumorigenicity Via circularRNA BRAF/microRNA-381-3p/Thrombospondin 2 in Glioma

Glioma is the most frequent primary intracranial tumor worldwide. Dexmedetomidine an efficient anesthetic used in surgery and has impact on glioma. Yet, mechanism of dexmedetomidine biological characteristics glioma cells unknown. Circ_BRAF, microRNA-381-3p thrombospondin 2 was detected via quantitative reverse transcription-polymer chine reaction. Proliferation detection conducted using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide colony formation assay. Transwell assay to determine cell migratory invasive capacities. The protein expression determined Western blot. Binding between microRNA- 381-3p circ_BRAF or validated dual-luciferase reporter. Role growth analyzed xenograft model mice. inhibited a dose-dependent manner. induced proliferation, migration invasion neutralized circ_ BRAF up regulation. Meanwhile, could facilitate binding with miR-381-3p. Besides, revealed promote tumourogenesis vivo circ_BRAF/ microRNA-381-3p/thrombospondin pathways. exposure expedited development circ_BRAF/microRNA-381-3p/thrombospondin axis, providing theoretical basis for therapy.
